LM11A-31 is an orally available, brain-penetrant small molecule that acts as a selective ligand for the p75 neurotrophin receptor (p75NTR). By modulating p75NTR signaling, it promotes neuronal survival pathways and inhibits apoptosis signaling. In preclinical models of Alzheimer’s disease and other neurodegenerative conditions, LM11A-31 has demonstrated neuroprotective effects by preventing synaptic loss, reducing tau phosphorylation and misfolding, decreasing microglial activation, and attenuating cholinergic neurite degeneration. It also blocks Aβ-induced neurodegeneration. Clinical trials have shown that LM11A-31 is safe and well-tolerated in patients with mild to moderate Alzheimer’s disease and may slow progression of synaptic degeneration as measured by biomarkers such as SNAP25 and NG[1][2][3][4][5].
