LMP766 is an orally bioavailable small molecule inhibitor of tumor necrosis factor-alpha (TNF-alpha) synthesis. Developed by Locus Pharmaceuticals using their proprietary computational fragment-based drug design platform, LMP766 was designed to treat inflammatory conditions such as Crohn's disease and rheumatoid arthritis. Unlike monoclonal antibody biologics that neutralize circulating TNF-alpha protein, LMP766 acts intracellularly to inhibit the production of TNF-alpha at the cellular level. Clinical development reached Phase 2 for the treatment of Crohn's disease; however, the program was ultimately terminated, and the developer, Locus Pharmaceuticals, ceased operations around 2010.
