Overview

LMT503 is an orally available small molecule drug developed by Lmito Therapeutics for the treatment of inflammatory bowel diseases such as Crohn disease and ulcerative colitis. It acts as an immunomodulator by modulating NADH quinone oxidoreductase (NQO1), which enhances NAD+ levels in macrophages. This leads to a shift in macrophage polarization from pro-inflammatory (M1-like) to anti-inflammatory (M2-like) phenotypes, resulting in reduced inflammation and potential amelioration of IBD symptoms. Preclinical studies have shown that LMT503 increases anti-inflammatory markers like IL-10 and Arg1 while decreasing pro-inflammatory cytokines such as TNF-α, iNOS, and IL-6. The drug is currently undergoing phase 1 clinical trials to assess its safety, tolerability, and pharmacokinetics in healthy volunteers[2][6][8].
