LN005

LN005 is an orally bioavailable, small molecule inhibitor of the fibroblast growth factor receptor (FGFR) family, specifically targeting FGFR1, FGFR2, and FGFR3. Developed by Shanghai Huiyong Pharmaceutical Co., Ltd., LN005 is designed to treat patients with advanced solid tumors characterized by FGFR genetic alterations, including gene fusions, mutations, or amplifications. The FGFR signaling pathway plays a critical role in cell proliferation, differentiation, and angiogenesis; its dysregulation is a known driver in several cancers such as non-small cell lung cancer (NSCLC), gastric cancer, and breast cancer. By competitively binding to the ATP-binding site of the FGFR kinase domain, LN005 inhibits the activation of downstream signaling cascades (e.g., MAPK/ERK and PI3K/AKT), thereby suppressing tumor growth and survival. It is currently undergoing early-phase clinical evaluation to assess safety, tolerability, and preliminary efficacy in refractory solid tumor populations.
