LN019 is a novel HER2-targeting affibody-drug conjugate (AfDC) designed for the treatment of HER2-overexpressing malignancies, including breast, ovarian, and gastric cancers. Utilizing a compact three-helix affibody scaffold, LN019 offers significant advantages over conventional monoclonal antibody-based conjugates, such as superior tissue penetration kinetics and picomolar binding affinity. Preclinical studies have demonstrated that LN019 exhibits potent dose-dependent cytotoxicity against HER2-positive cell lines and achieves rapid accumulation at tumor sites within minutes of administration. In xenograft models, LN019 has shown remarkable antitumor efficacy, inducing complete tumor regression at lower doses compared to trastuzumab deruxtecan (DS-8201). This therapeutic candidate is particularly promising for solid tumors where tissue penetration is a limiting factor for traditional antibody-drug conjugates.
