LN435a is a small molecule inhibitor of transforming growth factor beta receptor 1 (TGFBR1) developed by the Guangdong Provincial Hospital of Chinese Medicine. Derived from structural modifications of the natural alkaloid harmine, LN435a targets TGFBR1 to block the TGF-beta signaling pathway. It has demonstrated promising anti-tumor and anti-metastatic activity in breast cancer models, including patient-derived organoids (PDOs) and patient-derived tumor xenografts (PDX). By inhibiting TGFBR1, LN435a suppresses epithelial-mesenchymal transition (EMT) and cancer stem cell markers, reducing breast cancer extravasation and lung metastasis.
