Drug intelligence / Profile preview
LNA-ASO dual construct is a multitargeting antisense oligonucleotide (ASO) construct designed for the treatment of hepatitis B virus (HBV) infection [575]. Developed by the Vall d'Hebron Research Institute (VHIR) and the University of Barcelona, the construct consists of two biostable locked nucleic acid (LNA) ASOs linked together by a stimuli-responsive linker [575]. This linker is specifically cleaved by endogenous intracellular enzymes upon cellular entry, releasing both LNA-ASOs [575]. The released ASOs target hyper-conserved regions of the HBV genome, leading to the potent silencing of intracellular pregenomic RNA (pgRNA) and inhibition of viral replication [575].
