LNA043 is a modified recombinant version of the human angiopoietin-like 3 (ANGPTL3) protein developed as a novel disease-modifying osteoarthritis drug candidate. It acts as an agonist of ANGPTL3 and targets damaged cartilage by modulating several pathways involved in cartilage regeneration and homeostasis. Mechanistically, it promotes chondrogenesis and cartilage matrix synthesis by binding to the fibronectin receptor integrin α5β1 on mesenchymal stem cells and chondrocytes. Preclinical studies have shown that LNA043 induces hyaline articular cartilage regeneration, while clinical trials have demonstrated its safety profile and potential for regenerating damaged cartilage in patients with femoral articular cartilage lesions or knee osteoarthritis. The drug is administered via intra-articular injection[1][2][3][4]. Developed by Novartis and manufactured by Novartis.
