LNCaP + interleukin-2 + interferon-gamma is an allogeneic, whole-cell cancer vaccine developed for the treatment of prostate cancer. It is composed of the HLA-A2-matched LNCaP prostate cancer cell line, which has been genetically modified using retroviral transduction to secrete the immunostimulatory cytokines interleukin-2 (IL-2) and interferon-gamma (IFN-gamma). The vaccine is designed to present a broad spectrum of tumor-associated antigens (TAAs), including prostate-specific antigen (PSA), prostate-specific membrane antigen (PSMA), and prostatic acid phosphatase (PAP), to the host immune system. The locally produced cytokines act as molecular adjuvants: IL-2 facilitates the expansion and activation of T lymphocytes and natural killer (NK) cells, while IFN-gamma increases the expression of major histocompatibility complex (MHC) molecules and promotes a Th1-biased immune response. This combination strategy aims to overcome the immunosuppressive tumor microenvironment and induce a robust, systemic anti-tumor immune response. The product was primarily investigated in Phase I/II clinical trials for patients with hormone-refractory or castration-resistant prostate cancer.
