Overview

LNCB74 is an antibody-drug conjugate (ADC) that targets B7-H4, a protein highly expressed on multiple solid tumors, including breast, ovarian, endometrial, biliary tract, and non-small cell lung cancers. The drug consists of a B7-H4-specific antibody conjugated to the microtubule-disrupting payload monomethyl auristatin E (MMAE) via a glucuronidase-cleavable linker using LigaChem Biosciences’ ConjuAll technology. This site-specific linkage increases stability in circulation and enables selective release of the cytotoxic payload within tumor cells while minimizing off-target toxicity. LNCB74 incorporates an Fc region mutation to reduce uptake by immune cells expressing Fc receptors. Preclinical studies have demonstrated potent antitumor activity and favorable safety profiles in animal models. As of early 2025, LNCB74 is in Phase 1 clinical trials for advanced solid tumors.
