LNF2105 is an antibody-drug conjugate (ADC) targeting Nectin-4, developed by Shandong New Era Pharmaceutical (a subsidiary of Lunan Pharmaceutical Group). It consists of a humanized monoclonal antibody specific for Nectin-4 conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE). Nectin-4 is a cell surface adhesion molecule that is significantly overexpressed in various epithelial cancers, including urothelial, breast, and lung cancers, while maintaining low expression in normal tissues. Upon binding to Nectin-4, LNF2105 is internalized via endocytosis, and the cytotoxic payload is released through proteolytic cleavage of the linker. The released MMAE then binds to tubulin, inhibiting its polymerization and leading to G2/M phase cell cycle arrest and apoptosis. LNF2105 is currently in Phase 1 clinical development for the treatment of advanced solid tumors.
