**LNK01006** is a next-generation, orally available, brain-penetrant allosteric inhibitor of tyrosine kinase 2 (TYK2) that selectively targets the JH2 pseudokinase domain to modulate CNS-relevant inflammatory pathways, including IL-12/IL-23 signaling and type I interferons. Developed initially by Lynk Pharmaceuticals, it demonstrated high CNS exposure, potency, selectivity, and efficacy in preclinical models of multiple sclerosis (MS) by reducing pro-inflammatory Th1/Th17 T-cell activation and crossing the blood-brain barrier. Formation Bio acquired global rights (ex-Greater China) in 2025 via its subsidiary Bleecker Bio, planning Phase 1 trials in H1 2026 for neuroinflammatory and autoimmune diseases with central immune involvement.[1][2][4][7]
