LNK014 is an orally administered small molecule molecular glue degrader (MGD) designed to target NIMA-related kinase 7 (NEK7). Developed by Lynk Pharmaceuticals using its proprietary IsoNova protein degradation platform, LNK014 induces the proteasomal degradation of NEK7 by facilitating its interaction with the Cereblon (CRBN) E3 ubiquitin ligase complex. NEK7 is an essential scaffolding protein required for the assembly and activation of the NLRP3 inflammasome, which drives the maturation and release of pro-inflammatory cytokines such as IL-1β and IL-18. By selectively eliminating NEK7, LNK014 aims to block NLRP3 inflammasome activation and mitigate chronic inflammation. The drug is currently in preclinical development for the treatment of autoimmune and inflammatory diseases, including gout and inflammatory bowel disease (IBD).
