LNO-A01 is an antibody-drug conjugate (ADC) being developed by Linno Pharmaceuticals (also known as Lingnuo Pharmaceuticals) for the treatment of advanced solid tumors. The drug specifically targets Claudin 18.2 (CLDN18.2), a tight junction protein that is highly expressed in various gastrointestinal malignancies, including gastric, gastroesophageal junction, and pancreatic cancers, while maintaining limited expression in normal tissues. LNO-A01 utilizes a humanized anti-CLDN18.2 monoclonal antibody conjugated to a potent cytotoxic payload, likely a topoisomerase I inhibitor, via a proprietary site-specific conjugation platform. This technology is designed to produce a homogeneous drug product with a stable drug-to-antibody ratio (DAR), intended to enhance the therapeutic index and safety profile. As of early 2024, LNO-A01 is in the IND-enabling stage, with clinical development planned for both Australia and China.
