**LNP-C-3-80** is an experimental lipid nanoparticle mRNA cancer immunotherapy developed by Liangzhu Laboratory and Zhejiang University. It co-delivers mRNAs encoding an anti-CD3 single-chain variable fragment, a tumor-specific chimeric antigen receptor, and CD80 to engineer macrophages in vivo. The engineered macrophages are intended to form macrophage-T-cell-tumor-cell triads, providing tumor engagement, CD3-mediated T-cell activation, and CD80-mediated costimulation to enhance local antitumor immunity. In syngeneic mouse models of ovarian cancer and hepatocellular carcinoma, intraperitoneal or intravenous treatment produced marked tumor regression and reduced recurrence; no clinical development has been reported publicly.
