LNP-DNA with cancer-activated promoters is a proprietary gene therapy platform developed by Earli that utilizes lipid nanoparticles (LNPs) to deliver recombinant DNA constructs. These constructs are engineered with cancer-activated synthetic promoters that drive the selective expression of therapeutic proteins, such as Interleukin-2 (IL-2) and Interleukin-12 (IL-12), exclusively within tumor cells. This tumor-specific expression aims to maximize the therapeutic impact of immunotherapy agents while minimizing systemic toxicity and off-target effects in normal tissues. To enhance the systemic bioavailability and tumor accumulation of these LNPs, the platform can be combined with Nanobiotix’s Nanoprimer technology, which transiently occupies the mononuclear phagocyte system (MPS) to reduce hepatic clearance. The platform has demonstrated robust anti-tumor activity in syngeneic cancer models and is being optimized for extrahepatic delivery and improved tolerability by mitigating cGAS/STING-mediated inflammation.
