Overview

LNP-nCOV saRNA-02 is a self-amplifying RNA (saRNA) vaccine candidate encapsulated in lipid nanoparticles (LNPs), developed for the prevention of COVID-19 caused by SARS-CoV-2. The vaccine uses a modified saRNA platform designed to amplify antigen production within host cells, enabling robust immune responses at lower doses compared to conventional mRNA vaccines. The saRNA encodes the SARS-CoV-2 spike protein and includes viral replicase machinery (nonstructural proteins nsp1–4) that allows intracellular amplification of the RNA, resulting in high and prolonged antigen expression. This second-generation construct was specifically redesigned to dampen innate interferon responses and improve immunogenicity over earlier versions. Clinical trials have focused on evaluating safety and immunogenicity in healthy adults aged 18–45, with dosing regimens involving two intramuscular injections four weeks apart[1][2][9].
