Overview

LNR125 is a first-in-class fully humanized monoclonal antibody that targets and neutralizes interleukin-25 (IL-25), an upstream epithelial cytokine known as an "alarmin." IL-25 mediates Type 2 inflammation and is implicated in a range of atopic, allergic, fibrotic, viral infection and inflammatory diseases as well as certain cancers. By inhibiting IL-25 activity at the top of the inflammatory cascade, LNR125 aims to suppress multiple downstream cytokines simultaneously—potentially offering broader efficacy than therapies targeting individual downstream mediators. Preclinical studies have shown that LNR125 can inhibit allergic airway inflammation and augment anti-viral immunity in models of asthma and viral respiratory infection. The drug is being developed for indications including checkpoint inhibitor-induced pneumonitis (CIP), chronic urticaria, eosinophilic esophagitis, food hypersensitivity, gastroenteritis, rhinosinusitis; it has also been considered for asthma and atopic dermatitis[1][2][3][4][5].
