LNT 1 is a potent small molecule inhibitor of flap endonuclease 1 (FEN1), an enzyme involved in DNA replication and repair. It exhibits high selectivity for FEN1 (IC50 = 46.4 nM for hFEN1-336Δ) and is cytotoxic to SW620 colorectal cancer cells in vitro by inducing a DNA damage response. The compound has been used as a research tool to study synthetic genetic interactions of human FEN1 with DNA damage response genes and has demonstrated the ability to induce replication arrest, intra S-phase checkpoint activation, and apoptosis in glioma stem cells[1][3][8].
