Overview

LNZ101 is an investigational, preservative-free, single-use, once-daily ophthalmic eye drop formulation containing a combination of aceclidine and brimonidine. It is being developed for the treatment of presbyopia—the age-related loss of near vision—by LENZ Therapeutics. Aceclidine acts as a muscarinic acetylcholine receptor agonist to induce miosis (pupil constriction), creating a pinhole effect that improves near vision. Brimonidine is an alpha-2 adrenergic receptor agonist commonly used in glaucoma therapy; in this combination, it may help modulate pupil size and enhance or prolong the miotic effect while potentially reducing side effects such as redness. In Phase 2 and Phase 3 clinical trials (including the CLARITY program), LNZ101 demonstrated statistically significant improvements in near visual acuity without compromising distance vision, with rapid onset and effects lasting up to 10 hours. The drug was generally well tolerated with no serious treatment-related adverse events reported[2][5][6][9].
