LOAd703

LOAd703 (also referred to as LOKON003 in the context of its clinical trials) is an oncolytic gene therapy developed by Lokon Pharma. It consists of a replication-competent adenovirus (serotype 5/35) engineered to express two potent immunostimulatory membrane-bound ligands: CD40L (CD154) and 4-1BBL (CD137L). The therapy is designed for intratumoral administration to induce local tumor cell lysis and activate the immune system within the tumor microenvironment. By engaging CD40 on antigen-presenting cells and 4-1BB on T cells, LOAd703 promotes dendritic cell maturation and enhances the activation and expansion of tumor-specific T cells. It is primarily being investigated for the treatment of advanced solid tumors, including malignant melanoma and pancreatic cancer, often in combination with immune checkpoint inhibitors like atezolizumab to overcome resistance.
