LOAd732 is a novel immunostimulatory gene therapy based on an oncolytic adenovirus platform. It is engineered to express three key immunostimulatory transgenes—trimerized membrane-bound CD40 ligand (CD40L), 4-1BB ligand (4-1BBL), and interleukin-2 (IL-2). The mechanism of action involves selective infection and lysis of tumor cells, leading to direct tumor cell killing and the release of new viral particles. Additionally, the expressed transgenes activate dendritic cells (DCs) and stimulate antigen-specific T-cell and natural killer (NK) cell responses, even under immunosuppressive conditions commonly present in the tumor microenvironment. This dual action both destroys cancer cells directly and enhances anti-tumor immunity by overcoming local immune suppression. LOAd732 is being developed as a next-generation cancer immunotherapy[1][4][6].
