Lobenzarit is a small molecule immunomodulator and disease-modifying antirheumatic drug (DMARD) that was developed and approved in Japan for the treatment of rheumatoid arthritis. Unlike traditional nonsteroidal anti-inflammatory drugs (NSAIDs), lobenzarit does not inhibit prostaglandin or leukotriene biosynthesis; instead, it exerts its therapeutic effects by regulating the functions of T- and B-lymphocytes and suppressing the production of autoantibodies. It also possesses significant antioxidant properties, acting as a scavenger for oxygen-free radicals such as hydroxyl and superoxide radicals. Beyond arthritis, lobenzarit has been investigated for its potential in treating systemic lupus erythematosus, autoimmune liver disease, and inflammatory bowel disease (colitis), where it has shown the ability to inhibit the TLR4 and MAPK signaling pathways and restore mitochondrial homeostasis.
