Drug intelligence / Profile preview
Lock-Urelumab (also known as Pro-Urelumab or Lock-anti-4-1BB) is a tumor-selective, conditionally activated 4-1BB (CD137) agonist monoclonal antibody developed by PrecisemAb Biotech. It is engineered from the clinical anti-4-1BB antibody urelumab using PrecisemAb's proprietary Universal Antibody Lock technology. The antibody is masked with an autologous hinge-based lock connected via a matrix metalloproteinase-2/9 (MMP-2/9) cleavable peptide linker. This spatial hindrance reduces 4-1BB binding by approximately 400-fold in systemic circulation, preventing systemic T-cell co-stimulation and associated hepatotoxicity. Upon entering the protease-rich tumor microenvironment, MMP-2/9 cleaves the linker, releasing the lock and fully restoring 4-1BB agonist activity to stimulate localized antitumor immune responses. Lock-Urelumab is currently in preclinical development for the treatment of solid tumors.
