Lodotra is a modified-release (delayed-release) oral formulation of the corticosteroid prednisone. Developed by Mundipharma Research Limited and Horizon Pharma, it utilizes SkyePharma's proprietary GeoClock technology to provide a programmed lag time of approximately four hours after evening administration (typically at 10 p.m.). This timing ensures that the active glucocorticoid is released in the early morning hours (around 2 a.m.) to coincide with the peak of pro-inflammatory cytokines like IL-6, thereby more effectively managing morning stiffness and pain in patients with inflammatory conditions. It is indicated for the treatment of polymyalgia rheumatica, rheumatoid arthritis, and other inflammatory diseases.
