Overview
This investigational combination therapy, developed by the University of Pennsylvania, consists of lofexidine and pregabalin for the management of opioid withdrawal syndrome and the treatment of opioid use disorder. Lofexidine is an alpha-2 adrenergic receptor agonist that reduces the sympathetic overactivity associated with withdrawal, while pregabalin is a ligand for the alpha-2-delta subunit of voltage-gated calcium channels, which helps alleviate anxiety and pain. The combination is being evaluated in Phase 2 clinical trials (notably NCT04218240) to determine its efficacy in improving detoxification completion rates and facilitating the transition to extended-release naltrexone (XR-NTX) treatment.
