Overview

FP380 (lolamicin) is a systemic, Gram-negative-specific small molecule antibiotic developed by Flightpath Biosciences. It specifically inhibits the LolCDE complex, an ATP-binding cassette (ABC) transporter system responsible for the trafficking of lipoproteins from the inner membrane to the outer membrane in Gram-negative bacteria. By targeting this essential pathway, FP380 exhibits potent activity against pathogens such as *Escherichia coli*, *Klebsiella pneumoniae*, and *Enterobacter cloacae*. A key advantage of lolamicin is its high selectivity, which allows it to eliminate pathogenic bacteria while preserving the beneficial gut microbiome, potentially reducing the risk of secondary infections such as *Clostridioides difficile*. FP380 is currently being developed for the treatment of complicated urinary tract infections (cUTI) and sepsis, with human clinical trials anticipated to begin as early as 2026.
