Lolium perenne pollen depigmented polymerized extract is an allergen immunotherapy (AIT) product developed by LETI Pharma for the treatment of allergic rhinoconjunctivitis and allergic asthma triggered by perennial ryegrass pollen. The extract undergoes a specialized manufacturing process where the pollen proteins are depigmented to remove non-allergenic components (such as pigments and flavonoids) and subsequently polymerized with glutaraldehyde to form allergoids. This modification significantly reduces the extract's IgE-binding capacity, thereby decreasing the risk of systemic allergic reactions during administration, while maintaining the immunogenicity required to induce desensitization. The mechanism of action involves shifting the immune response from a Th2-dominated allergic profile to a Th1 and regulatory T-cell (Treg) response, characterized by the production of allergen-specific IgG4 blocking antibodies and the suppression of IgE-mediated mast cell and basophil activation.
