Overview

Lomitapide (AEGR-733) is a small molecule inhibitor of microsomal triglyceride-transfer protein (MTP), developed for the treatment of dyslipidemia, particularly in patients with homozygous familial hypercholesterolemia. By inhibiting MTP, lomitapide reduces the production of lipoproteins in the intestine and liver, thereby lowering blood levels of cholesterol and triglycerides. Atorvastatin is an HMG-CoA reductase inhibitor (statin) that decreases cholesterol synthesis in the liver to reduce LDL cholesterol and triglycerides while increasing HDL cholesterol. The combination has been studied for additive or synergistic effects on lipid lowering in patients with high cholesterol who do not reach target LDL-C levels with statins alone[1][2][6][7][8].
