Overview

This is a combination chemotherapy regimen consisting of three drugs: - **Lomustine** (CCNU), a nitrosourea alkylating agent that interferes with DNA and RNA function, causing cell death by damaging genetic material. It is cell cycle–phase nonspecific and used primarily for brain tumors and refractory Hodgkin's disease. - **Methotrexate**, an antimetabolite that inhibits dihydrofolate reductase, interfering with DNA synthesis, repair, and cellular replication. - **Mitomycin C**, an antineoplastic antibiotic that acts as an alkylating agent by cross-linking DNA strands to inhibit DNA synthesis. The combination has been studied notably in advanced non-small cell lung cancer (NSCLC) where it showed a regression rate of about 33%, median regression duration of approximately 4.9 months, and median survival around the same period. Toxicity includes cumulative myelosuppression as the most significant adverse effect but overall acceptable subjective toxicity[1][2][3][9].
