Overview

Lonafarnib + paclitaxel is a combination therapy consisting of two small molecule drugs used primarily in oncology research. Lonafarnib is a farnesyltransferase inhibitor that disrupts the post-translational modification of proteins involved in cell signaling and proliferation. Paclitaxel is a microtubule-stabilizing agent (taxane class) that inhibits cell division by promoting tubulin polymerization and preventing microtubule disassembly. The combination has demonstrated synergistic antiproliferative effects in preclinical models of ovarian cancer and clinical activity in patients with taxane-refractory or resistant metastatic non-small cell lung cancer (NSCLC). Mechanistically, the combination enhances mitotic arrest and apoptosis more than either agent alone, with evidence suggesting increased tubulin acetylation as part of its action[1][3][4][5].
