Overview

Lonapalene (RS-43179) is a small molecule drug that acts as a topically effective inhibitor of 5-lipoxygenase. It was developed for the treatment of psoriasis and demonstrated efficacy in double-blind clinical trials by reducing leukotriene B4 synthesis in psoriatic lesions. The mechanism of action involves inhibition of the enzyme 5-lipoxygenase, which plays a role in the inflammatory pathway by generating leukotrienes from arachidonic acid. Clinical studies showed significant improvement in psoriatic lesions treated with lonapalene compared to placebo. However, development was discontinued after Phase 3 trials[1][2][3][6][8].
