Overview

Lonasen Tape (DSP-5423P) is a transdermal patch formulation of blonanserin, an atypical antipsychotic developed by Sumitomo Pharma in collaboration with Nitto Denko. It is indicated for the treatment of schizophrenia. Blonanserin exerts its therapeutic effects primarily through high-affinity antagonism of dopamine D2 and D3 receptors, as well as serotonin 5-HT2A receptors. Compared to many other atypical antipsychotics, blonanserin has a higher affinity for D2 receptors than for 5-HT2A receptors and exhibits low affinity for adrenergic alpha-1, histamine H1, and muscarinic M1 receptors, which potentially reduces the incidence of side effects such as sedation and orthostatic hypotension. The transdermal formulation was developed to provide stable drug concentrations in the blood over a 24-hour period and to improve treatment adherence by offering a visible and easy-to-administer alternative to oral medications.
