**Loncastuximab tesirine + glofitamab** is a combination therapy under investigation for treatment of relapsed/refractory diffuse large B-cell lymphoma (DLBCL). Loncastuximab tesirine is an antibody-drug conjugate (ADC) targeting CD19, a B-cell antigen, with a humanized monoclonal IgG1 antibody conjugated to the cytotoxic SG3199 pyrrolobenzodiazepine (PBD) dimer via a protease-cleavable linker. It binds CD19, is internalized, and releases its cytotoxin to induce DNA crosslinks and cell death. Glofitamab is a bispecific monoclonal antibody that simultaneously binds CD20 on B cells and CD3 on T cells, recruiting and activating T cells for B-cell lysis. The combination leverages two distinct mechanisms—targeted cytotoxicity and immune-mediated killing—for potential synergy in aggressive B-cell lymphomas[1][2][3][5].
