Overview

Loncastuximab tesirine + rituximab is a combination therapy under investigation for the treatment of relapsed or refractory follicular lymphoma. Loncastuximab tesirine is an antibody–drug conjugate (ADC) consisting of a humanized anti-CD19 monoclonal antibody linked to a pyrrolobenzodiazepine dimer toxin (tesirine), which acts as a DNA cross-linking agent. Rituximab is a monoclonal antibody targeting CD20 on B cells, mediating cytotoxicity through immune mechanisms. The combination leverages the direct cytotoxic effect of loncastuximab tesirine and the immune-mediated effects of rituximab, with preclinical and clinical data suggesting synergistic activity between these agents[2][3][4][5][6]. ADC Therapeutics developed loncastuximab tesirine, which is FDA-approved as monotherapy for relapsed/refractory large B-cell lymphoma; its use in combination with rituximab for follicular lymphoma remains investigational[6].
