Overview

This is a combination therapy regimen consisting of four agents: loncastuximab tesirine (an anti-CD19 antibody-drug conjugate), rituximab (an anti-CD20 monoclonal antibody), bendamustine (a nitrogen mustard alkylating agent), and cytarabine (a nucleoside analog). The combination is being investigated for the treatment of various B-cell lymphomas, including relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma. Loncastuximab tesirine was approved by the FDA in April 2021 for relapsed/refractory DLBCL after at least two lines of treatment. The combination of rituximab, bendamustine, and cytarabine (R-BAC) has shown efficacy in mantle cell lymphoma (MCL) with favorable toxicity profiles.
