This is a combination chemotherapy regimen consisting of three agents: - **Lonidamine** is a small molecule that inhibits aerobic glycolysis in tumor cells by targeting mitochondrial hexokinase and other metabolic pathways, thereby sensitizing cancer cells to cytotoxic agents. - **Mitomycin-C** is an antitumor antibiotic that acts as an alkylating agent, cross-linking DNA and inhibiting DNA synthesis. - **Vinblastine** is a vinca alkaloid that disrupts microtubule formation during cell division (mitosis), leading to cell cycle arrest and apoptosis. The combination has been studied as salvage therapy for advanced breast cancer, particularly in patients previously treated with anthracycline-based regimens. Clinical studies have shown partial and complete responses with manageable toxicity profiles. The main toxicities are related to lonidamine (myalgia, abdominal cramps, reversible deafness) while hematological toxicity was uncommon[1].
