Lonigutamab ugodotin (W0101) is an antibody-drug conjugate (ADC) under development for the treatment of advanced or metastatic solid tumors. The drug consists of a monoclonal antibody (hz208F2-4) that specifically targets the insulin-like growth factor type 1 receptor (IGF-1R), which is overexpressed in various tumor types. This antibody is conjugated to a highly potent cytotoxic auristatin derivative via a novel linker. Upon binding to IGF-1R on tumor cells, the ADC is internalized and releases its cytotoxic payload, leading to selective killing of cancer cells while sparing normal tissues. Preclinical studies have shown that W0101 induces receptor-dependent cell cytotoxicity and significant tumor regression in models with high IGF-1R expression. A first-in-human Phase I/II clinical trial is ongoing to evaluate safety and efficacy in patients with advanced or metastatic solid tumors[6][2][3].
