Lonoctocog alfa is a recombinant human coagulation factor VIII (FVIII) used for the treatment and prevention of bleeding in patients with hemophilia A. It is a single-chain FVIII molecule with a truncated B-domain, allowing covalent linkage between the heavy and light chains, which improves its stability and affinity for von Willebrand factor (VWF). This enhanced VWF binding stabilizes FVIII in circulation and protects it from degradation. Lonoctocog alfa replaces the missing or deficient endogenous FVIII in hemophilia A patients, thereby restoring effective hemostasis by enabling normal blood clotting. It is indicated for both on-demand treatment of bleeding episodes and prophylaxis to prevent bleeds across all age groups[1][3][5][6][7].
