Overview

This entry refers to a specific combination of antiretroviral agents—lopinavir, atazanavir, and ritonavir—evaluated in clinical research conducted by The HIV Netherlands Australia Thailand Research Collaboration (HIV-NAT). Specifically, these drugs were used as boosted protease inhibitor (PI) components in pharmacokinetic studies (such as NCT00475215) to assess their interaction with tenofovir disoproxil fumarate (TDF) in HIV-infected Thai children. Lopinavir and atazanavir are HIV-1 protease inhibitors that prevent the cleavage of gag-pol polyproteins, resulting in the production of immature, non-infectious virus particles. Ritonavir is used in sub-therapeutic doses as a pharmacokinetic enhancer (booster) because it potently inhibits the CYP3A4 enzyme, thereby increasing the plasma concentrations and half-lives of other protease inhibitors like lopinavir and atazanavir.
