Lorecivivint is a small molecule investigational drug developed as a potential disease-modifying osteoarthritis drug (DMOAD), primarily for knee osteoarthritis. It acts as an intra-articular inhibitor of CDC-like kinase 2 (CLK2) and dual-specificity tyrosine phosphorylation-regulated kinase 1A (DYRK1A), modulating the Wnt signaling pathway. By inhibiting these intranuclear kinases, lorecivivint improves chondrocyte health and function while reducing inflammation. Preclinical and clinical studies have shown that it preserves cartilage structure, increases joint space width, reduces pain and improves function in patients with moderate to severe knee osteoarthritis.
