Overview

Lorexys is a fixed-dose combination of bupropion and trazodone being developed by S1 Biopharma for the treatment of hypoactive sexual desire disorder (HSDD) in premenopausal women. Bupropion is a norepinephrine-dopamine reuptake inhibitor (NDRI) that enhances sexual desire by increasing dopamine and norepinephrine levels in the brain's reward pathways. Trazodone is a serotonin antagonist and reuptake inhibitor (SARI) that, at low doses, acts primarily as a 5-HT2A receptor antagonist, which can help mitigate the anxiety and inhibitory signals that suppress sexual desire. The combination aims to restore the balance between excitatory (dopamine/norepinephrine) and inhibitory (serotonin) neurotransmitter systems involved in sexual response.
