Lorigerlimab is an investigational, bispecific monoclonal antibody designed to target both programmed cell death protein 1 (PD-1) and cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) on tumor-infiltrating lymphocytes. By binding to these immune checkpoint proteins, lorigerlimab inhibits their mediated downregulation of T-cell activation and proliferation, thereby enhancing antitumor immune responses. The drug is based on MacroGenics' DART (Dual-Affinity ReTargeting) platform and is being developed primarily for the treatment of metastatic castration-resistant prostate cancer (mCRPC), with additional studies in solid tumors. Lorigerlimab has demonstrated a manageable safety profile and encouraging antitumor activity in early-phase clinical trials.
