This is a multi-drug combination regimen consisting of eight targeted anticancer agents: lorlatinib (ALK/ROS1 inhibitor), encorafenib and binimetinib (BRAF and MEK inhibitors), talazoparib (PARP inhibitor), sacituzumab govitecan (Trop-2-directed antibody-drug conjugate with SN-38 topoisomerase I inhibitor payload), axitinib (VEGF receptor inhibitor), fam-trastuzumab deruxtecan-nxki (HER2-directed antibody-drug conjugate with deruxtecan topoisomerase I inhibitor payload), and enfortumab vedotin (Nectin-4-directed antibody-drug conjugate with monomethyl auristatin E payload). It targets multiple oncogenic pathways including ALK fusion, BRAF mutation, DNA repair via PARP, angiogenesis via VEGF, and overexpressed surface antigens (Trop-2, HER2, Nectin-4) in advanced solid tumors. Being evaluated in the StrataPATH precision medicine trial (NCT05097599) for patients with actionable molecular alterations matching these drugs' indications, primarily in non-small cell lung cancer, melanoma, breast cancer, and other solid tumors.[10]
