Overview

Lormetazepam is a short- to intermediate-acting benzodiazepine derivative and a temazepam analogue. It acts as a hypnotic primarily indicated for the short-term treatment of moderate-to-severe insomnia. Lormetazepam exerts its effects by binding to the regulatory site between the α and γ subunits of the gamma-aminobutyric acid (GABA) A receptor complex in the central nervous system. This positive allosteric modulation enhances GABAergic neurotransmission by increasing chloride ion influx into neurons and hyperpolarizing them—resulting in sedative-hypnotic effects as well as anxiolytic, anticonvulsant, muscle relaxant properties. It was patented in 1961 and introduced into medical use in 1980[1][3][6].
