Overview

Lorukafusp alfa is an intravenously administered immunocytokine, a recombinant fusion protein comprising a humanized 14.18 anti-GD2 monoclonal antibody linked to interleukin-2 (IL-2). Its mechanism of action involves targeting disialoganglioside GD2 on the surface of tumor cells, leading to antibody-dependent cell cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Additionally, it activates natural killer (NK) cells and T cells through binding to the interleukin-2 receptor. This dual mechanism contributes to its anti-tumor activity. The drug has been developed by several entities including Apeiron Biologics, Merck KGaA, National Cancer Institute (USA), University of Wisconsin-Madison, Lexigen Pharmaceuticals, and Bristol Myers Squibb Co.
