Overview

Lorvotuzumab mertansine is an investigational antibody-drug conjugate (ADC) designed for the treatment of CD56-positive cancers, such as small-cell lung cancer, ovarian cancer, multiple myeloma, leukemia, and Merkel cell carcinoma. The drug consists of a humanized monoclonal antibody (lorvotuzumab or huN901) that specifically targets Neural cell adhesion molecule 1 (NCAM1/CD56), conjugated via a cleavable disulfide linker to DM1 (mertansine), a potent microtubule-disrupting cytotoxic agent derived from maytansine. Upon binding to CD56-expressing tumor cells, the ADC is internalized and releases DM1 inside the cell, leading to disruption of microtubules and induction of apoptosis. Lorvotuzumab mertansine has shown promising results in early-phase clinical trials for several malignancies with high CD56 expression and has been granted orphan drug status for Merkel cell carcinoma[1][2][3][4][5][6][8].
