Overview
Losartan + aspirin is a combination of two repurposed small-molecule drugs being evaluated as adjunct agents in combination immunotherapy regimens. Losartan is an angiotensin II receptor blocker (AT1 receptor antagonist) that modulates the tumor microenvironment by depleting the extracellular matrix, reducing tumor hypoxia, and improving drug delivery. Aspirin (acetylsalicylic acid) is a cyclooxygenase (COX) inhibitor with anti-inflammatory and antiplatelet properties that provides immunomodulatory effects. In the context of BreakBio Corp's BreakVax trial, these agents are used to enhance the efficacy of personalized neoantigen-based immunotherapy, checkpoint inhibitors (pembrolizumab, ipilimumab), and chemotherapy in patients with advanced solid tumors, including triple-negative breast cancer and glioblastoma.
