Overview

Lotamilast is a small molecule, selective phosphodiesterase 4 (PDE4) inhibitor developed primarily for the topical treatment of atopic dermatitis. By inhibiting PDE4, lotamilast increases intracellular cyclic AMP levels, which leads to reduced production of proinflammatory cytokines and chemokines, thereby exerting anti-inflammatory and antipruritic effects. The drug was originally discovered by Eisai and later developed by Dermavant Sciences (a subsidiary of Organon). Clinical studies have shown statistically significant improvements in atopic dermatitis symptoms with topical application[1][2][3][4][5][6][7].
