Overview

low affinity IL-3z.CAR is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target CD123 (interleukin-3 receptor alpha), a protein frequently overexpressed in acute myeloid leukemia (AML) and blastic plasmacytoid dendritic cell neoplasm (BPDCN). Unlike conventional CARs that utilize antibody-derived single-chain variable fragments (scFvs) for antigen recognition, this construct employs a mutated, low-affinity version of the natural IL-3 cytokine as its extracellular binding domain. This design strategy aims to improve the therapeutic window by selectively activating T cells against malignant blasts with high CD123 density while sparing healthy hematopoietic stem and progenitor cells (HSPCs) that express lower levels of the antigen. By reducing off-tumor toxicity, specifically myeloablation, this approach seeks to provide a safer alternative to high-affinity CD123-targeted therapies.
